When conducting an Espionage realm action, must the type of espionage be the same in every province?

As far as I know, yes.

I wonder though if you are able to do different espionage actions as a realm action, if you are only affecting a single province. This is how rule holding works.

The above is incorrect.

Realm action Rule holdsing only allow a single holding type to be built.

Similarly, a realm Espionage would only allow one type of espionage to occur, as far as I know.

It need not be, but all actions need to be thematically linked; i.e. as long as all actions taken have to do with bringing the Grey Duke to justice they can be of different types - counterespionage, assassination, whatever.
